What are Nano-Secondaries®?

Nano-Secondaries® are a novel class of secondary antibodies for higher resolution & cleaner images. They consist of Nanobodies/ VHHs that bind to primary antibodies with high affinity in a species and subtype-specific manner. Nano-Secondaries are conjugated to Alexa Fluor® dyes.

What:

Dr. Astrid Sitte and Dr. Rebecca Northeast will provide best practices for immunoprecipitation (IP) and immunofluorescent (IF) experiments.

In this webinar, we discuss the principles of IP and protocol optimization. First, we provide recommendations on selecting the appropriate antibody or Nano-body. Next, we discuss different bead matrices and antibody immobilization methods. The practical steps of an IP are then outlined from sample preparation through the elution step with a focus on the major parameters that determine the experiment’s success.

 IF visualizes multiple proteins of interest in the same sample to investigate distribution and colocalization of proteins of interest in different disease and experimental states. We provide an overview of our workflow, taking you step-by-step through each procedure and outlining the different options available for optimization. We discuss how to choose the correct primary and secondary antibodies for your experiments, and how directly conjugated primary antibodies can be used to enhance your results. Furthermore, we describe how Nano-secondary antibodies can be used for super-resolution microscopy and provide excellent resolution and reproducibility.  Common troubleshooting points and solutions will then be addressed to close the talk.
