Background Information
FBXO38, also known as HMN2D, MOKA, and SP329, is a protein that plays a significant role in the regulation of immune responses, particularly in the context of cancer and autoimmune diseases. FBXO38 has been identified as an E3 ubiquitin ligase that mediates the degradation of PD-1 (Programmed cell death protein 1), a key immune checkpoint protein expressed on the surface of T cells. The internalization, ubiquitination, and proteasome degradation of PD-1 are facilitated by FBXO38, which links PD-1 to Lys48, leading to its poly-ubiquitination and subsequent degradation (PMID: 30487606).
