New nuclear Actin-Chromobody® plasmid

The nuclear Actin-Chromobody® plasmid
The nuclear Actin-Chromobody® (nAC) is a novel tool to visualize and analyze endogenous nuclear actin dynamics in living cells. The lab of Prof. Grosse, University of Marburg, Germany has added a nuclear localization sequence (NLS) to the original Actin chromobody plasmid.
Upon transformation, the cells express an anti-Actin VHH, which is fused to tagGFP and NLS (for an excellent schematic see figure 1A from Plessner et al. 2015). The intracellular expressed nAC is then imported into the nucleus and binds to Actin. In their publication Plessner et al. demonstrated that the transfection of nAC did not affect the abundance of endogenous nuclear actin. Therefore, they have used the nAC to analyze the nuclear actin dynamics during cell spreading and showed that Integrin signaling can induce the polymerization of nuclear actin.
