Background

U-937 cytoplasmic extract was prepared from cell cultures of the human histiotic lymphoma U-937 cell line. The U-937 cell line was derived from malignant cells of a pleural effusion of a 37-year old male patient. These cells are of myeloid lineage and differentiate into a mature macrophage phenotype in response to various stimuli. U-937 cells produce and secrete large quantities of cytokines and chemokines either spontaneously or in response to treatment with soluble stimuli, such as TNFα. In addition, IFNβ and IFNϒ stimulation has been shown to induce the expression of chemokine receptors on the cell surface of U-937 cells and results in increased production of interleukins. Because U-937 cells are one of only a few human cell lines with characteristic monocytic properties, they are commonly used in research as in vitro models for monocyte/macrophage differentiation.

U-937 cytoplasmic extract is supplied in Dilution Buffer (20 mM Hepes (pH 7.9), 100 mM KCl, 1 mM MgCl2, 20% glycerol, 0.5 mM PMSF and 0.5 mM DTT).
