Background Information

Tissue factor pathway inhibitor (TFPI) is an anticoagulant protein produced primarily by the endothelium and megakaryocytes and alternative mRNA splicing generates two main forms, TFPIα and TFPIβ (PMID: 26149025, PMID: 16246254). TFPIα is a soluble form of TFPI. It is a ~46 kDa glycoprotein and consists of an acidic N-terminal regionfollowed by three tandem, Kunitz-type protease inhibitor domains and a basic C-terminal region. Based on protein mass, TFPIβ (28 kDa) is considerably smaller than TFPIα (36 kDa), but both migrate with the same apparent molecular mass (46 kDa) on sodium dodecylsulfatepolyacrylamide gel electrophoresis (SDS-PAGE), suggesting a difference in post-translational modifications (PMID: 16246254). TFPI associated with LDLis 34 kDa and lacks a substantial portion of the carboxy terminus including at least a portion of the third Kunitz domain. TFPI associated with HDL is 41 kDa and is in part a mixed disulphide complex between TFPI and apolipoprotein All (PMID: 8979119). TFPI exerts its anticoagulant effects by inhibiting TF-FVIIa in a manner that is dependent on its inhibition of factor Xa (FXa) (PMID: 2927510).
