KE20017 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The rat CXCL2 ELISA kit is to be used to detect and quantify protein levels of endogenous rat CXCL2. The assay recognizes rat CXCL2. An antibody specific for rat CXCL2 has been pre-coated onto the microwells. The rat CXCL2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for rat CXCL2 is added to detect the captured rat CXCL2 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Chemokine (C-X-C motif) ligand 2 (CXCL2) is a small cytokine belonging to the CXC chemokine family that is also called macrophage inflammatory protein 2-alpha (MIP2-alpha), Growth-regulated protein beta (Gro-beta) and Gro oncogene-2 (Gro-2). This chemokine is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2. During inflammation, the level of CXCL2 increases and is involved in the release of opioid peptides from leukocytes. Down-regulation of CXCL2 could reduce the recruitment of polymorphonuclear cells, which are key players in the clearance of L. monocytogenes during infection.
