Product Information

KE00045 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The INS ELISA kit is to be used to detect and quantify protein levels of endogenous INS. The assay recognizes human INS. An antibody specific for INS has been pre-coated onto the microwells. The INS protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for INS is added to detect the captured INS protein. For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Insulin is a peptide hormone, produced by beta cells of the pancreas, and is central to regulating carbohydrate and fat metabolism in the body. It participates in glucose utilization, protein synthesis and in the formation and storage of neutral lipids. Insulin is synthesized as a precursor molecule, proinsulin, which is processed prior to secretion. A- and B-peptides are joined together by a disulfide bond to form insulin, while the central portion of the precursor molecule is cleaved and released as the C-peptide. Defects in insulin results in type 1 diabetes mellitus.
