Product Information

KE00060 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The IDH1 ELISA kit is to be used to detect and quantify protein levels of endogenous IDH1. The assay recognizes human IDH1. An antibody specific for IDH1 has been pre-coated onto the microwells. The IDH1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for IDH1 is added to detect the captured IDH1 protein. For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

IDH1, also named as PICD and IDP, belongs to the isocitrate and isopropylmalate dehydrogenases family. IDH1 may be a tumor suppressor that, when mutationally inactivated, contributes to tumorigenesis in part through induction of the HIF-1 pathway(PMID: 19359588). It is a common feature of a major subset of primary human brain cancers. IDH1 mutation is always heterozygotic and IDH1 functions as a dimer, theoretically there will be 25% each wild type and mutant homodimers and 50% heterodimers present in the tumor cells(PMID:21079649).
