KE00393 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Cyclin E1 ELISA kit is to be used to detect and quantify protein levels of endogenous human Cyclin E1. The assay recognizes human Cyclin E1. An antibody specific for human Cyclin E1 has been pre-coated onto the microwells. The human Cyclin E1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Cyclin E1 is added to detect the captured human Cyclin E1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Cyclin E1 (CCNE1) is a member of the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. CCNE1, an essential cyclin activating cyclin-dependent kinase 2 (CDK2), regulates the G1-S phase transition of the mammalian cell division cycle.The transition from the quiescent state of the cell cycle into the phase of DNA synthesis is thought to be regulated by E-type cyclins Cyclin E1 (CCNE1) and Cyclin E2 (CCNE2) in complex with CDK2. During G1/S phase, cyclin dependent kinases (CDKs) (CDK4/6) is activated by cyclin D1, D2, or D3 to phosphorylate Rb, followed by phosphorylation by CDK2 in complex with cyclin E1 or cyclin E2. Cyclin E1 is highly related to cyclin E2, and both cyclin E1 and cyclin E2 are estrogen target genes that can facilitate anti-estrogen resistance and can be highly expressed in breast cancer. Amplified expressed cyclin E1 is a well-known tumorigenic factor and prognostic biomarker in several malignancies, such as osteosarcoma .
