Product Information

Proteintech's PK10031 (H&E staining kit) can be used for tissue and cell staining. Cell nuclei can be stained blue while cytoplasm can be stained red. The formulas of the reagents in the kit have been optimized to avoid using toxic chemicals, e.g. mercury or methanol. Staining with the H&E Staining Kit is fast and easy. 

The product is provided in a ready-to-use format. No further dilution is required. Each milliliter of this product can stain ~10 slides.

Ambient. Stable for one year.

Usage

For FFPE samples:

1. Immerse slides in xylene for 10 minutes. Repeat in fresh xylene again for another 10 minutes. Then rehydrate the slides by sequentially incubating in 100%, 95%, 80%, and 60% ethanol solutions for 5 minutes each. Finally wash the slides with distilled water for 2 minutes.

2. Apply Mayer's Hematoxylin Solution from the kit onto the tissue slides and make sure the tissues are fully covered. After staining for 1-2 minutes, wash the slides with distilled water. (If staining is too dark, the staining could be lightened by immersing the slides in 1% HCl ethanol solution). Apply bluing reagent from the kit to the slides for 2 minutes. Once the sample turns blue, wash the slides briefly with distilled water 3 times. 

3. Apply Eosin staining solution from the kit to the slides and make sure the tissues are fully covered. Stain for 2-5 minutes. Wash the slides with distilled water after staining.

4. Dehydrate the slides with gradient ethanol followed by clearing, mounting and observation. 

For frozen samples:

1. Wash the slides with distilled water, apply Mayer's Hematoxylin Solution from the kit onto the tissue slides and make sure the tissues are fully covered. After staining for 1-2 minutes, wash the slides with distilled water. (If staining is too dark, the staining could be lightened by immersing the slides in 1% HCl ethanol solution). Apply bluing reagent from the kit to the slides. Blue for 2 minutes. Once the sample turns blue, wash the slides briefly with distilled water for 3 times. 

2. Apply Eosin staining solution from the kit to the slides and make sure the tissues are fully covered. Stain for 2-5 minutes. Wash the slides with distilled water after staining.

3. Dehydrate the slides with gradient ethanol followed by clearing, mounting and observation.

Cautions

1. Stain with 1-2 samples to make sure the staining achieves optimal results. 

2. Wear a lab coat and gloves during operation.
