Background Information

DYRK (Dual-specificity tyrosine phosphorylation-regulated kinase 2) is the only member of the Dual-specificity tYrosine phosphorylation-Regulated Kinase (DYRK) family that functions as a kinase activity-independent scaffold for an E3 ubiquitin ligase complex (PMID: 33376136). DYRK2 is involved in cell survival, development, differentiation, gene transcription, proteasomal regulation, and microtubule formation. DYRK2 is a key regulator of cell cycle and apoptosis in response to DNA damage via phosphorylation of p53. DYRK2 has two isoforms and is mostly localized in the cytoplasm; however, it accumulates in the nucleus under genotoxic stress conditions (PMID: 31505048, 37886981).
