Product Information

KE00435 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ROCK1 ELISA kit is to be used to detect and quantify protein levels of endogenous human ROCK1. The assay recognizes human ROCK1. An antibody specific for human ROCK1 has been pre-coated onto the microwells. The human ROCK1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ROCK1 is added to detect the captured human ROCK1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Rho-associated protein kinase 1 (ROCK1) is a serine/threonine protein kinase that was identified as a RhoGTP-binding protein. To date, two isoforms encoded by two different genes of ROCK (ROCK1 and ROCK2) have been investigated. ROCK1 and ROCK2 have been shown to fulfill major roles in carcinogenesis. These two proteins share an overall sequence similarity in their kinase domains of 92%, and at the amino-acid level, a similarity of 65%. ROCK1 is involved in the pathogenesis of metabolic-related diseases, including hypertension, arteriosclerosis, Alzheimer's disease, and diabetes.
