  IHC staining of human tonsillitis using 17476-1-AP

  Immunohistochemical analysis of paraffin-embedded human tonsillitis tissue slide using 17476-1-AP (CCR5 antibody) at dilution of 1:800 (under 10x lens. Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  FC experiment of K-562 using 17476-1-AP

  1X10^6 K-562 cells were stained with 0.2ug CCR5 antibody (17476-1-AP, red) and control antibody (blue). Fixed with 90% MeOH blocked with 3% BSA (30 min). Alexa Fluor 488-conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) with dilution 1:1000.

Geprüfte Anwendungen

• Erfolgreiche Detektion in IHC: humanes Tonsillitisgewebe; Hinweis: Antigendemaskierung mit TE-Puffer pH 9,0 empfohlen. (*) Wahlweise kann die Antigendemaskierung auch mit Citratpuffer pH 6,0 erfolgen.
• Erfolgreiche Detektion in FC: K-562-Zellen

Empfohlene Verdünnung

• Immunhistochemie (IHC): IHC : 1:400-1:1600
• Durchflusszytometrie (FC): FC : 0.20 ug per 10^6 cells in a 100 µl suspension

It is recommended that this reagent should be titrated in each testing system to obtain optimal results.

Hintergrundinformationen

CCR5 (also known as CD195) is a member of the beta chemokine receptor family. This protein is expressed by T cells and macrophages and is known to be an important co-receptor for macrophage-tropic viruses, including HIV, to enter host cells. The natural chemokine ligands that bind to CCR5 include MIP-1-alpha, MIP-1-beta, MCP-2, and RANTES.
