  FC experiment of human PBMCs using CL488-69002

  1X10^6 PMA+ionomycin-stimulated and Monensin Solution treated human PBMCs were intracellularly stained with 0.25 ug CoraLite® Plus 488 Anti-Human TNF Alpha (CL488-69002, Clone:11H8A5), and 0.25 ug Mouse IgG1 Isotype Control (CL488-66360, Clone: T1F8D3F10), and 0.25 ug CoraLite® Plus 647 Anti-Human CD3 (UCHT1) (CL647-65151, Clone: UCHT1). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  1X10^6 PMA, LPS, and Brefeldin A treated THP-1 cells were intracellularly stained with 0.25 ug CoraLite® Plus 488 Anti-Human TNF Alpha (CL488-69002, Clone:11H8A5) (red), or 0.25 ug Control Antibody. Cells were fixed with 4% PFA and permeabilized with Intracellular Staining Permeabilization Wash Buffer.

  FC experiment of NK92 using CL488-69002

  1X10^6 PMA, ionomycin, and Brefeldin ANK92 were intracellularly stained with 0.4 ug CoraLite® Plus 488 Anti-Human TNF Alpha (CL488-69002, Clone:11H8A5) (red), or 0.4 ug x. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

TNF, as also known as TNF-alpha, or cachectin, is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. It is expressed as a 26 kDa membrane bound protein and is then cleaved by TNF-alpha converting enzyme (TACE) to release the soluble 17 kDa monomer, which forms homotrimers in circulation. It is produced chiefly by activated macrophages, although it can be produced by many other cell types such as CD4+ lymphocytes, NK cells, neutrophils, mast cells, eosinophils, and neurons. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, INS resistance, and cancer.
