Applications Validated by Active Motif: IP: 10 ul per IP WB*: 1:500 - 1:2,000 dilution *Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot. |

Background Information
MED1 (Mediator complex subunit 1, TRIP2, Thyroid receptor-interacting protein 2) is a component of the Mediator complex, a coactivator involved in the regulated transcription of most genes transcribed by RNA polymerase II. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional pre-initiation complex with RNA polymerase II and the general transcription factors. MED1 is involved in supporting hematopoietic stem and/or progenitor cell function.
