Background Information
AIB3 (Amplified in Breast Cancer 3, Nuclear Receptor Coactivator 6, NCoA6) is a chromatin-associated protein that interacts with nuclear hormone receptors (antibodies, kits and proteins) to modify their activity as transcriptional activators. AIB3 / NCoA6 was originally isolated as a ligand-dependent nuclear receptor interacting protein. It is a co-activator that directly binds nuclear receptors and stimulates their transcriptional activity in a hormone-dependent fashion. AIB3 is a multifunctional co-activator necessary for transcriptional activation of a wide spectrum of target genes. AIB3 is involved in growth, development, wound healing and maintenance of energy homeostasis. It can enhance the transcriptional activation of a variety of transcription factors and also associates with a number of important regulators of transcription including CBP and p300. AIB3 / NCoA6 is a component of at least three distinct MLL complexes, suggesting that AIB3 plays a fundamental role in transcriptional activation by modulating chromatin structure through histone methylation. AIB3 may also be involved in the co-activation of the NF-kappa-B pathway. It is amplified and overexpressed in breast, colon and lung cancers. AIB3 function is essential in mammalian development as AIB3 null embryos do not survive. Additionally, AIB null spontaneously undergo apoptosis, indicating that AIB3 has anti-apoptotic and pro-survival functions.
